Mid 30's, a *little bit* of cash to spend, no idea where to start or what to pick up, probably lean toward Marvel slightly as opposed to DC just based on my limited awareness of the product. Looking for suggestions and shit. Go!

Pick up the Essentials books. They are usually pretty cheap (between $15 and $20) and you get a lot of bang for your buck. You can find any of them on Amazon, usually for less than cover price. I have essential Fantastic Four volumes 1-8 (which reprints issues 1-183, plus annuals 1-11, plus some tie-ins, to give you some perspective), Avengers 1-8, Thor 1-5, Spider-Man 1-5, and some random ones, like Adam Warlock, Dr Strange, Marvel 2-in-1 and so on. Really worth the money.
I also recommend the Marvel Omnibus editions. I have the John Byrne Fantastic Four Omnibus Volume 1 (Which is about fifty comics on glossy stock under a hardcover) for $125 retail. I got it for $60 on eBay. You should definitely pick up Frank Miller's Daredevil, some of the Marvel Visionaries stuff, and you can't go wrong with some of the Older X-Men stuff.
My recommended buying/reading list: The Dark Knight Returns Batman: Year One Watchmen Frank Miller's Daredevil Sin City Cerebus Infinity Gauntlet The Trial of Galactus Superman/Batman: Generations Jack Kirby's Fourth World Walt Simonson's Thor
Some of the other guys can clue you in on stuff that's come out in the last ten years, like Civil War and AvX. I don't buy anything new except some occasional stuff from IDW.

Honestly, I'd focus on third party stuff unless you just REALLY wanna stick with Marvel and DC. There are a lot of really cool books put out by Image and others that aren't bogged down with 50 years of continuity. If you wanna stick with the Big Two, I can't argue Shlocko's list. If you are open to other stuff:
The Walking Dead
Invincible - A really fun series that is a bit of Satire of superhero books, but has some great serious storylines.
Bone - Haven't read yet, but heard good things.
Ex Machina - A superhero becomes Mayor of New York after saving one of the Twin Towers on 911.
Ireedeemable - What if Superman turned heel on us?

Honestly, you can't go wrong with Punisher: MAX series written by Garth Ennis.
The first 7 books or so are my all time favorite series of comics. They're for adults. So there's no "Motherfriggin killer!" or guys getting hit with bullets off the page. This is based in real life, with gritty story-lines and violence. It's absolutely fantastic.
You'd start at Punisher: MAX - The Beginning, and go until about Girls In White Dresses.

Supreme Power was also great. It was a retelling of the Squadron Supreme (basically the JLA in the Marvel Universe) done by the MAX line.

I got into comics later on in life like you, mid 20s, some great stand alones to pick up are Watchmen, Dark Knight Rises and I'm surprised it's not been mentioned but Batman: The Killing Joke. Also, you mentioned liking Buffy in another thread, honestly the Buffy comics are very hit and miss but Angel: After The Fall 1-14 are EXCELLANT and have a great story to them, there's also a one (two) shot called Angel: Boys and their Toys that may be my favorite Buffy related comic. It has Angel, Spike and Groo going to a comic book convention to get a mystical sword in an auction but a bunch of demons are there and they use the same spell Ethan Rayne used in that Halloween episode turning everybody into their costumes including Spike, who is dressed as Angel. Also while their not canon like the other comics, Spike:Asylum and Spike:Shadow Puppets are great reads.

If you are sticking with mainstream Marvel/DC
The essentials/Showcase series will give you the most bang for your buck.
The essentials are B&W though, might be a detriment. Although I do like Walkng Dead.
But if you go the Trade paperback/Graphic Novel route until you find a book worthy of getting single issues.
